We are pleased to announce the release of version 5.1.1 of the open-source Fluorescence Lifetime fitting and visualisation tool FLIMfit, providing a number of new features and performance enhancements. Download version 5.1.1 here.
Please note that this version of FLIMfit uses Maximum Likelihood fitting by default. This is the best choice for TCSPC datasets..
A guide to using FLIMfit for analysing multiphoton FLIM data with tutorial videos is available in a recent paper, Conway, JRW, Warren, SC & Timpson, P, Context-dependent intravital imaging of therapeutic response using intramolecular FRET biosensors, Methods (2017).

New features
- New segmentation manager with new freehand tool options and improved performance
- Estimate IRF from a mono-exponential reference using an exponentially modified gaussian model.
This can produce a high quality IRF for TCSPC detectors without secondary peaks or significant afterpulsing
- Add support for HydraHarp and TimeHarp data files
- Add support for data acquired during bidirectional scan
- Where possible determine repetition rate from data (currently ptu/pt3 files)
- Significantly improved loading times for some data formats
- Added option to display smoothed data in the decay tab. When off, this significantly reduces the time to switch between datasets.
This option does not affect smoothing during data fitting.
- Improved performance selecting/deselecting images to fit
- Update to Bioformats 5.4
Other Enhancements
- Add option to plot weighted or unweighted values in plotter
- Visual enhacements to plotter
- Enable plotting by image number
- Allow setting default Gate Max value
- Improved support for older MacOS X versions (10.9.x)
Bug fixes
- Allow t0 shift map estimation from any loaded dataset (rather than first only)
- Fix crash when fitting t0 and offset locally

This version is built with Bio-Formats 5.2 (specifically version 5.2.2) which, among other improvements, includes support for Becker & Hickl .spc FIFO data and performance improvements for loading large .sdt FLIM files.

We are pleased to announce the release of version 4.11.1 of the open-source Fluorescence Lifetime fitting and visualisation tool FLIMfit. Download version 4.11.2 here.
Updates/Improvements
- FLIMfit is now capable of handling some multi-dimensional file-sets. For example, if you have a real-time time-series of z-stacks, with one z-stack in each of several files, it can now be successfully loaded and processed.
- The “Files->Load FLIM Data” menu item now allows the user to select multiple files.
- The “Files->Load FLIM Dataset” menu item has been renamed, for clarity, to “Load FLIM Data from a Directory”.
- The maximum number of segmented regions has been increased to 4095.
- Another waitbar has been added to make the workflow clearer when using large datasets.
- A method for estimating a Gaussian IRF from single-exponential reference data has been added
- Loading Picoquant files created on MT200 systems is now supported
Bug Fixes
- An issue with loading polarisation resolved IRFs, with different time points to the data, has been addressed
- An issue with the loading of time-varying backgrounds has been addressed.
- An issue loading time gated data with legacy file names has been addressed
